INCLUSIVE FLOW CYTOMETER SAMPLE PREPARATION WORKFLOWS

The DRG is meant for direct acquisition on flow cytometers. A list of validated cytometers for use with the DRG and tested parameter settings can be found below.

The acquisition parameters should be used as directions and may require optimization with your specific application and cytometer. For use on cytometers other than those validated, please reach out to your regional Field Application Scientist for assistance.

The DRG is a disposable volume adaptor designed to sit on top the Laminar Wash™ Plate forming physical walls around each well to increase the total holding volume of the plate for direct acquisition of samples for flow cytometry and other downstream applications. The DRG can be acquired stably on closed flow cytometer up to 2 -3 hours (for more observations, please check with Curiox Application Support.

Waste containment: regular lab requirements for labware

CENTRIFUGE-LESS CELL PROCESSING FOR FLOW CYTOMETRY ASSAYS

Traditional centrifuge-based preparation of cells are time consuming, labor intensive, waste expensive reagents, and stress cells. There is significant cell loss and incomplete washing resulting in problems with gating and cell segmentation.

Laminar washing delivers better cell retention and cleaner, clearer data

When performing cell dissociation in tumor bearing tissues, the tissue debris and enzymatic-induced fragments are primary factors affecting cell recovery for tumor infiltrating lymphocytes. For more invasive and immunologically challenging models, like B16F10-inoculated mice, recovery of limited numbers of TILs reduces chances for accurate characterization. The results shown here were demonstrated with an unchallenged mouse sample and the Laminar Wash technology. Laminar washing (with 1,000,000 tumor infiltrate cells) reveals increased viable cell count and reduced debris effects by laminar washing. The cleaner sample preparation increases lymphocyte recovery up to 90.6% as compared to centrifuge-harsh washing of 10.5%.

HOW LAMINAR WASH™ WORKS

The power of Curiox’s laminar cell washing technology lies in the unique design of the 96-sample plate. Each plate contains an array of hydrophilic spots that suspension cells can rest on via gravity. The material surrounding each spot is highly hydrophobic, acting as an impenetrable wall that separates your samples.

Upon entry into the instrument, two nozzles descend onto each side of every spot, one to dispense wash buffer and the other to aspirate the buffer and debris away.

Each spot can wash as many as 10 million cells down to a single cell without the mechanical stress and cell losses associated with centrifugation.
